In resistant insects phosphine slowly reacts with some component of the membrane which retards its entry but may slowly oxidize it to hypophosphite. Since this site is not the toxic lesion no harm is done. In susceptible insects phosphine is transported unchanged into the internal tissues where it disrupts sensitive redox systems thus generating free radicals. The biochemical processes involved in this unique resistance mechanism have yet to be elucidated. Despite the technical difficulties which have hindered biochemical studies of insecticide resistance in stored-product insects, an increasing stream of such studies is now being reported. I believe that this is important since the potential for development of resistance in these insects is enormous and their economic and social importance is considerable, especially in developing countries. It is to be hoped that the emerging use of molecular biological techniques such as those used on aphids at Rothamsted and on mosquitoes in France and California, can be usefully employed to elucidate resistance mechanisms in stored-product insects.
